Reaction(IUBMB)
(2E,6E)-farnesyl diphosphate = (1R,4R,5S)-(-)-guaia-6,10(14)-diene + diphosphate [RN:R11830]
Reaction(KEGG)
R11830
Substrate
(2E,6E)-farnesyl diphosphate [CPD:C00448]
Product
(1R,4R,5S)-(-)-guaia-6,10(14)-diene [CPD:C21685];
diphosphate [CPD:C00013]
Comment
The original enzyme (STC5) from the fungus Fusarium fujikuroi is inactive because of a critically naturally occuring mutation that leads to an asparagine to lysine exchange in the NSE (Asn-Ser-Glu) triad, a highly conserved motif of type I terpene cyclases. Sequence correction by site-directed mutagenesis (K288N) restores activity.
