#=GF CC stimulating peptide precursors, which are generally up to 50
#=GF CC amino acid residues long. In all the members of this family, the
#=GF CC leader sequence is cleaved after two conserved glycine residues;
#=GF CC thus the leader sequence is of the double- glycine type [2].
#=GF CC Competence stimulating peptides (CSP) are small (less than 25
#=GF CC amino acid residues) cationic peptides. The N-terminal amino
#=GF CC acid residue is negatively charged, either glutamate or
#=GF CC aspartate. The C-terminal end is positively charged. The third
#=GF CC residue is also positively charged: a highly conserved arginine
#=GF CC [2]. A few COMC proteins and their precursors (not included in
#=GF CC this family) do not fully follow the above description. In
#=GF CC particular: the leader sequence in the CSP precursor from
#=GF CC Streptococcus sanguis NCTC 7863 Swiss:O33758 is not of the
#=GF CC double-glycine type; the CSP from Streptococcus gordonii NCTC
#=GF CC 3165 Swiss:O33645 does not have a negatively charged N-terminus
#=GF CC residue and has a lysine instead of arginine at the third
#=GF CC position. Functionally, CSP act as pheromones, stimulating
#=GF CC competence for genetic transformation in streptococci. In
#=GF CC streptococci, the (CSP mediated) competence response requires
#=GF CC exponential cell growth at a critical density, a relatively
#=GF CC simple requirement when compared to the stationary-phase
#=GF CC requirement of Haemophilus, or the late-logarithmic- phase of
#=GF CC Bacillus [1]. All bacteria induced to competence by a particular
#=GF CC CSP are said to belong to the same pherotype, because each CSP
#=GF CC is recognised by a specific receptor (the signalling domain of a
#=GF CC histidine kinase ComD). Pherotypes are not necessarily
#=GF CC species-specific. In addition, an organism may change pherotype.
#=GF CC There are two possible mechanisms for pherotype switching:
#=GF CC horizontal gene transfer, and accumulation of point mutations.
#=GF CC The biological significance of pherotypes and pherotype
#=GF CC switching is not definitively determined. Pherotype switching
#=GF CC occurs frequently enough in naturally competent streptococci to
#=GF CC suggest that it may be an important contributor to genetic
#=GF CC exchange between different bacterial species [2]. The family
#=GF CC Antibacterial16, streptolysins from group A streptococci, has
#=GF CC been merged into this family.
